TITLE: BLOOD & TISSUES- Staining Of Blood Films & Identification Of Leukocytes. The erythrocytes are slightly visible, but the leukocytes are excessively pale, just about invisible and we will not see anything inside them. To be able to obser ve and recognize the diverse kinds of leukocyte, we must dirt them. For this purpose, normally Giemsa turd and Wright stain are used. Giemsa stain is a mixture of dyes, based on methylene blue (basic dye) and eosin ( acidic dye). Giemsa stain is a classical blood film stain for peripheral blood smears and bone pump specimens. The principle of Giemsa stain is the basic dye stained the acidic sectionalisation of the burden of the cell and the acidic dye stained the basic part of the cytoplasm of the cell. Therefore, the Giemsa stain allows clear observations to discern types of leukocytes.
